| Further information: | The plasmid was constructed by isolating the Tet-On element and multicloning site from pT-REx-MCS and ligating it into the NdeI/XbaI opened pCDNA3-zeo vector. The plasmid is intended for tet-on expression in cells where neomycin selection does not work, for example Jurkat cells. There is uncertainty about the presence of a second enhancer in the SV40 early promoter. The absence of a second enhancer in this promoter may cause bacterial leakage expression of the Tn5 neomycin resistance gene. |
