Further information: | The plasmid was constructed by introducing S73C, A115C, A143C and P183C mutations into the S. coelicolor LPMO10C coding sequence of pCXP34h-ScLPMO10C via site-directed mutagenesis. The S. coelicolor LPMO10C coding sequence was codon-optimised for E. coli. The mutations introduced into the S. coelicolor LPMO10C coding sequence improve the stability of the encoded protein. The LPMO10C protein can be recovered from the periplasmic space. The nucleotide sequence of the wild type S. coelicolor LPMO10C coding sequence corresponds with the EMBL Nucleotide Sequence Database accession number AL939108.1. Other name of the plasmid is ScLPMO10C_A143C_P183C_S73C_A115C. |
