Further information: The plasmid was constructed by introducing L636Q, L637M, K639R, T640V, V641R and A642N mutations into the A. caldus SuSy coding sequence of pCXP34h-SuSyAc via site-directed mutagenesis.
The plasmid is meant for the constitutive expression of the His-tagged A. caldus mutant in E. coli BL21 DE3. This mutant has an increased affinity for UDP glucose.
The nucleotide sequence of the A. caldus SuSy coding sequence was codon optimised for expression in E. coli.
The nucleotide sequence of the wild type A. caldus SuSy cDNA corresponds with the EMBL Nucleotide Sequence Database accession number KP284426.1.
