| Further information: | To construct this vector, the A. nidulans ans1 sequence was transferred from pFB6-An2 to pDJB1 as an isolated 3.5 kb EcoRI fragment. The 1.8 kb PvuII-PstI fragment of the ans1 sequence enhances the efficiency of transformation of this plasmid. This plasmid contains a 1.9 kb genomic HindIII-BglII fragment of Neurospora crassa, including the pyr4 gene coding for orotidine 5'-phosphate decarboxylase; this enzyme can also be expressed in Escherichia coli, using part of the Neurospora sequence as the promoter. Since the nucleotide sequence of the ans1 insert and of the pyr4 insert could not be completely traced back, there is uncertainty as to the cutting frequency of the restriction enzymes, except for the sites that were analysed in the authenticity test. |
