Further information: The plasmid was constructed by restriction digest of pEASY-FLIRT-mCYLD-LAH-3K and pEASY-FLIRT-mCYLD-RAH-R321A with NotI/SalI, isolating the fragments containing the mouse CYLD homology arms of both parental vectors and ligating them together.
This is a knock-in construct in order to generate a MALT1-resistant CYLD mouse. The MALT1 cleavage site was eliminated via a R321A mutation. The mouse CYLD coding sequence also contains a NaeI site, introduced via a silent mutation.
The plasmid contains two bacteriophage P1 Cre recombinase target sites (loxP sites) for excision of the cassette by Cre-recombinase.
pEASY-FLIRT contains a testis-specific Flp expression cassette (Ace-Flpe) aimed at deleting the frt-flanked NeoR gene in male gonads of chimeric mice.
