Further information: The plasmid was constructed by isolating the FLAG-tagged human CARD14 mutant from pCDNA3.1-FLAG-hCARD14-E142K and cloning it into the EcoRV/XbaI opened pEF6/Myc-HisA vector.
The human CARD14 mutant was recloned from pcDNA3.1 to pEF6 to allow experiments with different levels of NF-kappaB/AP-1 inductions. The hCMV-IE promoter is responsive to both of these transcription factors, while the hEF1α promoter is less variable.
The nucleotide sequence of the wild type human CARD14 coding sequence corresponds with the EMBL Nucleotide Sequence Database accession number AF322642.1.
