Cloned DNA: Human chemokine (C-C motif) ligand 11 (CCL11, Eotaxin-1)
Myc epitope; C-terminal
Histidine tag (His-tag); C-terminal
Promoter: Human elongation factor 1α promoter (EF1α)
Phage T7 gene 10 promoter (T7g10)
Simian virus 40 early promoter (SV40 early)
Synthetic prokaryotic EM7 promoter
Escherichia coli lac operon promoter
Ribosome
binding site:
-
Terminator: Bovine growth hormone polyadenylation signal (BGH polyA)
Simian virus 40 polyadenylation signal (SV40 polyA)
Selection marker: Ampicillin (amp)
Blasticidin (bsd)
Replicon: Escherichia coli plasmid pMB1 origin
Phage f1 origin
Simian virus 40 bidirectional origin (SV40)
Host range: Escherichia coli; preferably recombination-deficient and endonuclease A deficient strains
Mammalian cells; SV40 permissive cells
Parental clone: pEF6/Myc-HisA
Further information: The plasmid was created by isolating the human CCL11 CDS from spleen cDNA via PCR and cloning it as a BamHI/XbaI fragment into pEF6/Myc-HisA.
The human CCL11 CDS includes a termination codon, hence the Myc epitope and His-tag will not be translated.
The nucleotide sequence of the human CCL11 cDNA corresponds with the Genbank accession number NM_002986.2.
EMBL Accession number: NM_002986.2, view at GenBank

Sequence detail:
Primers used to amplify the human CCL11 CDS:

Forward: 5' AGGTGGATCCACC.ATG.AAG.GTC.TCC.GCA.GCA.CTT
                BamHI     ***

Reverse: 5' ATATTCTAGA.TTA.TGG.CTT.TGG.AGT.TGG.AGA.T
                XbaI   +++

***: start codon.
+++: terminatin codon.
Punctuation indicates reading frame.
