Further information: The plasmid was created by introducing a multiple cloning site into the pFAJ1706 vector as a HindIII-BamHI fragment.
This plasmid is a stable RK2-derived cloning vector that can be used for the study of genes in a wide background of gram-negative bacteria.
The plasmid RK2 trfA gene encodes two polypeptide products P382 and P285 of 382 and 285 amino acids respectively, using a different initiation codon within the same reading frame. The resulting protein is essential for vegetative replication.
The plasmid carries the Rhizobium sp. NGR234 spsAB postsegregational killer locus to ensure stable maintenance of the plasmid.
The multiple cloning site of the plasmid is protected against read-through transcriptional activity of vector sequences by the trpA transcriptional terminator.
