Further information: | The plasmid was constructed by inserting the SacI/HindIII fragment of pRK5-B1EC2S2, containing the 3' part of the human TGFB1 coding sequence, between the SacI and HindIII sites of the polylinker of pGEM-11Zf(+). The human TGFB1 coding sequence contains C223S and C225S mutations (C2S2) and has an engineered starting codon. These mutations result in expression of human TGFB1 that is largely biologically active. The plasmid can be used for the production of single-stranded RNA probes (riboprobes). When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
