| Further information: | The plasmid was constructed as follows: 1) the 926 bp mouse CASP-11 promoter fragment plus the first 25 nucleotides of the mouse CASP-11 mRNA were picked up with a mouse genome walker kit (Clontech); 2) the PCR amplicon was blunted and phosphorylated (Klenow/T4-kinase) and 3) subcloned in the SmaI linearized, dephosphorylated pGL3-Basic vector using T4 DNA ligase. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT726959.1. The nucleotide sequences of the mouse CASP-11 promoter fragment and mRNA fragment correspond with the EMBL Nucleotide Sequence Database accession number AF539416.1, except for two nucleotides in the promoter sequence: the 'A' nucleotides at EMBL positions 489 and 677 are 'G' nucleotides. Other name of the plasmid is pGL3-casp11p-FL-WT. |
