| Further information: | The plasmid was constructed by inserting a 822 bp fragment, containing the Nicotiana sylvestris glutathione peroxidase homologue (GPX) cDNA, into the EcoRI opened pBluescriptIIKS- vector. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of the pBluescriptIIKS- vector corresponds with the Genbank accession number X52329.1 and adapted to the restriction enzyme pattern analysis results. The following difference is observable: the two BssHII sites are missing. The nucleotide sequence of the N. sylvestris GPX cDNA corresponds with the Genbank accession number X60219.1. Other name of the plasmid is pGluper. |
