Further information: The plasmid was constructed by creating an in-frame deletion of 234 bp in the Y. enterocolitica sycT gene of pIM154 via ClaI/HindIII digestion, Klenow treatment and religation, resulting in pIM161. A SalI/XbaI fragment of pIM161 containing the sycTΔ16-94 allele was subcloned into the corresponding sites of the suicide vector pKNG101, resulting in mutator plasmid pIM164. The sycTΔ16-94 allele was finally introduced in the wild-type strain Y. enterocolitica E40 (pYV40) by double recombination with pIM164, to obtain Y. enterocolitica E40 [pIM402].
Other names of the plasmid are pYV40 sycT and pYV40 sycTΔ16-94.
