| Further information: | The plasmid was constructed by creating an out of frame deletion of 428 bp after codon 135 of Y. enterocolitica yopT of pIM157 by NhiI/EcoRI digestion, Klenow treatment and religation, resulting in pIM166. An XbaI/EcoRI fragment of pIM166 encoding the yopT135 and 94 codons of sycT was subcloned in the XbaI/EcoRI opened pBluescriptIIKS- vector, resulting in pIMB199. A SalI/XbaI fragment of pIMB199 encoding yopT135 and sycT94 was subcloned into the SalI/XbaI opened pKNG101 suicide vector, resulting in the pIML200 mutator plasmid. pIML200 was mobilised in Y. enterocolitica E40 by conjugation, and the wild type gene was replaced by the yopT135 allele by double recombination. Other names of the plasmid are pYV40 yopT and pYV40 yopT135. |
