| Further information: | The plasmid was constructed by inserting the 1.1 kb BamHI-EcoRI fragment, containing the P. freudenreichii cobA gene, between the BamHI and EcoRI sites of pUC19. The BamHI site has been lost (personal communication of C. Roessner). The plasmid can be used for a fluorescense-based method for selection of recombinant plasmids. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727342.1. The nucleotide sequence of the cobA insert corresponds with the EMBL Nucleotide Sequence Database accession number U13043.2. |
