| Further information: | To make this plasmid, pIVHA1 was BamHI opened, filled in (Klenow), and ligated with a linker coding for termination codons in all three reading frames ('CT TAG T TAA C TAA G'). The SalI fragment, containing the HA1si gene, codes for a secreted haemagglutinin via an mRNA similar to that of the HA1 gene, except for the termination codon inserted at the BamHI site. Only two amino acids were added by the linker insertion: Leu (amino acid 187; identical to the wild type sequence) and Ser. Other name of the plasmid is pSRHA1mSTOP.. |
