Genecorner:

-

Cloned DNA: Phage fd gene 3 (g3)
Promoter: Phage λ major leftward promoter (λ PL)
Ribosome
binding site:
Ribosome binding site (RBS) of the phage fd gene 3
Terminator: -
Selection marker: Kanamycin (kan)
Replicon: Escherichia coli plasmid ColE1 origin
Host range: Escherichia coli; use strains with a cI function
Parental clone: pPLa2311
Further information: Gene 3 of fd phage was first cloned as an AluI-AluI fragment into the HindII site of pUR222; then the cloned gene was inserted in pPLa2311 as a PstI-EcoRI fragment.
No repression of P1 is needed; the fd-3 protein is not toxic and can be constitutively expressed.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727597.1.
