| Further information: | The plasmid was constructed by cloning a fragment of the phage T7 genome, containing the T7 lysozyme coding sequence, into the BamHI-opened pACYC184 vector. This plasmid is intended for co-transformation with a bacterial expression vector in E. coli BL21(DE3) (LMBP 1455). The phage T7 DNA is present in opposite orientation compared to the tet promoter, consequently pLysS (Silent) supplies a low level of phage T7 lysozyme. This is sufficient to stabilize many target plasmids and yet allow high levels of target protein to be produced upon induction of T7 RNA polymerase. |
