Further information: | The plasmid was constructed by PCR amplifying the 5' region of human PPID1 from IMAGE clone 3908102 and the 3' region from IMAGE clone 4852968. A 5' VSV-tag and a 3' FLAG epitope were added during PCR amplification. The human PPID1 coding sequence was fused, and cloned into the pMSCVpuro vector. The plasmid contains the mouse stem cell PCMV (PCC4-cell-passaged myeloproliferative sarcoma virus) virus long terminal repeat. The LTR drives high-level constitutive expression of human PIDD1 in stem cells and other mammalian cell lines. The plasmid contains also the extended retroviral packaging signal, Ψ+, which promotes high-titer production of replication-incompetent infectious viral particles after introduction into a packaging cell line. Other name of the plasmid is pMSCV-PIDDisof1-Flag. |
