Further information: | The plasmid was constructed by replacing the EcoRI-XhoI fragment from pMaSVT2 (containing the SV40-E promoter) by the EcoRI-XhoI fragment from pGEMIL6gIL2f1. Hence, the SV40 large T-antigen is placed under control of the human interleukin 6 promoter. This plasmid differs from pMaSVTHIL61 in the length of the 5' flanking region of the hIL6 promoter. pMaSVTHIL62 has 12 nucleotides more and 3 BamHI sites, instead of 2. After mutagenesis, use mutS strains for primary transformation (e.g. sup(-) strain WK6mutS, sup(+) strain BMH71-18mutS); for segregation of possible mutants: sup(-) strains (e.g. WK6). Name of the phasmid mentioned in Vanhoenacker et al. (1994) is pMaHIL6SVT. The nucleotide sequence of the genomic hIL6 DNA corresponds with the EMBL Nucleotide Sequence Database accession number AC073072.11. However, the C nucleotide at promoter position -174 (polymorphism mutant) was changed into a G nucleotide (wild type). |
