| Further information: | pPLcATnHIFNB plasmids are all structural variants of the prototype ribosome binding site and were constructed to test the relevance of this feature for efficient initiation of translation. pPLcAT141HIFNB is a human IFNB expression plasmid. This plasmid belongs to the pPLcATnHIFNB series. This plasmid contains a 52 bp AluI fragment from human IFNB (coordinates 5 to 57) before the SD sequence. The distance between SD and ATG codon is 8 bases (filled in XbaI site) which abolishes completely the IFNB synthesis. All other pPLcATnHIFNB plasmids have 4 nucleotides between SD and ATG codon. The nucleotide sequence of the human IFNβ DNA corresponds with the EMBL Nucleotide Sequence Database accession number V00546.1. |
