Further information: The plasmid was constructed by cloning the VSV-ADIP sequence into Xho/EcoRV opened pPpT4alpha-VSV-hMALT1Cd100-His vector, upstream of the human MALT1 fragment.
Adiponectin is a multimerization domain (Mashalidis et al., 2013) meant to increase in vitro activity in low salt conditions.
The plasmid expresses the minimal paracaspase domain of human MALT1, in frame of the S. cerevisiae ppMF, wich means that Pichia induced by methanol will secrete the protein construct.
The expressed protein can be purification via the C-terminal His-tag. The N-terminal VSV tag allows verification that the purified product is intact.
The nucleotide sequence of the human MALT1 coding sequence corresponds with the EMBL Nucleotide Sequence Database accession number AF130356.2.
