Further information: | This plasmid allows the high-level production of a recombinant single chain Fv fragment of the mouse IgG1 antibody against the BCL1 antigen. The recombinant product contains a 6xHis-tag at the carboxy terminus. The promoter/operator element T5 N25/O2 consists of the highly efficient E. coli phage T5 N25 promoter and two lac operator sequences (Bujard et al. 1987). The presence of the lacI(q) repressor gene is necessary to control the activity of the promoter. The synthetic ribosome binding site RBSII is described in Stüber et al. (1990). The actual sequences of the ribosome binding site RBSII in front of the pelB signal sequence as well as the junction between pelB and scFv have been experimentally verified.
