| Further information: | pSCGAL1mBAX-T was derived from pSCGAL1mBAX-L by replacing the Ecl136II-MluI fragment (blunted with Klenow DNA polymerase), containing the S. cerevisiae leucine 2 coding sequence (LEU2) and the S. cerevisiae iso-1-cytochrome C terminator (CYC1), by the BamHI fragment (blunted with Klenow DNA polymerase) of YDp-W, containing the TRP1 coding sequence. pSCGAL1mBAX-T is an integrative expression plasmid for the mouse Bax-α protein in S. cerevisiae. After PmlI (PmaCI) linearisation in its TRP1 element, single-copy genomical integration leads to stable expression. The BamHI site at position 3942 could not be experimentally confirmed. The nucleotide sequence of the mBAX cDNA corresponds with the EMBL Nucleotide Sequence Database accession number L22472.1. Other names of the plasmid are YIpUTT-muBax and pSCGALmBAX3. |
