Further information: The plasmid was constructed by inserting the Candida albicans O-acetylhomoserine O-acetylserine sulfhydrylase cDNA, allele 1 (CaMET15-1), provided by SfiI and NotI linkers between the SfiI and NotI sites of pSCGAL10-SN.
Expression of the cloned cDNA is controlled by the regulatable GAL10/CYC1 hybrid promoter from Saccharomyces cerevisiae and the iso-1-cytochrome C (CYC1) and 2μ FLP terminators. The hybrid promoter sequence consists of the upstream activation sequence (UAS-G) of the galactokinase gene (GAL10), the GAL10 promoter and the 5' untranslated leader sequence of the cytochrome C1 (CYC1) gene.
The defective LEU2 gene (leu2-d) is poorly expressed, because there is only 30 bp of upstream sequence left in this plasmid; the expression is probably controlled by a promoter within the 2μ sequences.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727323.1.
The nucleotide sequence of the CaMET15-1 gene corresponds with the Genbank accession number AF188273.1.
Other names of the plasmid are pSCGAL10CaMET15-3 and pSCGALMET15-3.
