Further information: | This plasmid was constructed as follows: 1) The intermediate plasmid pSCGCMFE6L was constructed by ligating 3 fragments : a) the XbaI/EcoRI vector fragment of pEMBLyex4, b) the EcoRI/SalI fragment of pEMBLyex4, containing the Saccharomyces cerevisiae 2μ origin and c) the SalI/XbaI fragment of pSCGAL1MFE6L, containing the prepro secretion signal sequence (ppMF) of the S. cerevisiae α-mating factor 1 gene (MFα1) fused to cDNA encoding the light chain of the mouse anti-hPLAP monoclonal antibody E6(IgG2b,κ), E6L. 2) The EcoRI fragment of pSCUDT2MFE6H, containing the prepro secretion signal sequence (ppMF) of the S. cerevisiae α-mating factor 1 gene (MFα1) fused to cDNA encoding the heavy chain of the mouse anti-hPLAP monoclonal antibody E6(IgG2b,κ), E6H, was then inserted into the EcoRI opened pSCGCMFE6L vector. The plasmid is used for expression of the heavy chain of the mouse anti-hPLAP monoclonal antibody E6(IgG2b,κ): E6H. |
