Further information: The plasmid was constructed as follows: 1) a cDNA expression library from a Nicotiana plumbaginifolia (tobacco) heterotrophic cell suspension culture was constructed in pUC18; 2) a clone synthesizing a SOD enzyme with characteristic properties of a tobacco chloroplastic iron superoxide dismutase cDNA (FeSOD), resistant to KCN and sensitive to H2O2, was selected.
The inserted FeSOD fragment is fused to lacZα and expressed under control of the lacZ promoter. The first 25 amino acid codons of FeSOD are missing.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727068.1.
The nucleotide sequence of the FeSOD cDNA corresponds with the Genbank accession number M55909.1.
