| Further information: | The plasmid was constructed as follows: 1) a cDNA library from a Nicotiana plumbaginifolia (tobacco) cell suspension culture was constructed in pUC18; 2) an oligo of 26 nucleotides, matching a highly conserved region of the C-terminal part of the coding sequence of the tobacco cytosolic copper/zinc superoxide dismutase cDNA (CuZnSOD), was used as a probe to screen; 3) the clone with the largest cDNA insert was selected. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727067.1. The nucleotide sequence of CuZnSOD cDNA corresponds with the Genbank accession number X55974.1. |
