| Further information: | The plasmid was constructed as follows: into the unique BamHI site of pSP64, a RsaI-RsaI fragment (containing the Saccharomyces cerevisiae galactokinase promoter (GAL1)) was inserted via BamHI linkers. The BamHI site at the 3' end of the GAL1 promoter was lost during construction; the sequence in this region is not exactly known. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. |
