| Further information: | This plasmid was constructed as follows: 1) First, the unique BamHI site of pSV23S was opened, filled in with Klenow DNA polymerase and ligated; 2) Then, the StuI-EcoRI fragment from this intermediate vector, containing the splice signal of SV40 as well as the polyA signals, was inserted into the SmaI and EcoRI sites of pSP64Mx. This plasmid contains the mouse Mx promoter including 19 nucleotides downstream from the TATA-box, ending at position -1 which is the middle of the lost PvuII site. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727544.1. Other name of the plasmid is pMxSA. |
