Further information: | The plasmid was constructed by inserting the first EcoRI fragment of the EcoRI linked hIL4 cDNA gene into the unique EcoRI site of pSP65, in the antisense orientation relative to the SP6 promoter. The first EcoRI linker was placed approx. 60 nucleotides upstream from the start codon of the hIL4 cDNA; the natural EcoRI site is located 100 nucleotides upstream from the termination codon of the hIL4 cDNA. This plasmid is useful for preparing antisense RNA probes. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The hIL4 cDNA was not sequenced but assumed to be identical to the sequence reported by Yokota et al. (1986). |
