Further information: The synthetic suppressor tRNA gene (SupF) carries an EcoRI site at both termini. The length of this synthetic gene is 209 nucleotides (including the EcoRI sites).
The synthetic suppressor gene was phosphorylated at the 5' ends and then joined to an ColE1-amp(R) vector after the latter had been digested by EcoRI.
Expression of the supF gene is controlled by its own promoter.
