Further information: | Construction: SV40 part ranging from the EcoRI site clockwise to nucleotide 989; junction between VP1 and polyA signal with the splicing signal of the small t-antigen via the BclI site (SV40 coordinate 2770) and the MboI site (SV40 coordinate 4100); junction between the splicing signal of the small t-antigen and SV40 early promoter: via the MboI site (SV40 coordinate 4710) and a HindIII linker with a HindIII-PvuII fragment containing the promoter; junction between SV40 early promoter and pAT153: via PvuII site and filled- in AvaI site from pAT153 (pAT153 coordinate 1425).(see PhD thesis p130) Contains the SV40 small t-antigen splicing signal. Since prokaryotic part is derived from pAT153, there are no 'poison' sequences present. Designed for transient expression in COS cells and constitutive expression in CHO cells (cotransfection with DHFR plasmid). Expression sites: a unique HindIII site. It is presumably possible to clone into the AvrII and/or StuI sites for expression, but this has not been tested. Other name of the plasmid is pSV2d. |
