Further information: This plasmid was constructed as follows: the human TNF cDNA from pATHTNF was inserted into the unique SalI site of pSV23S after SalI linkers (CGTCGACG) were ligated to the PstI sites of the human TNF cDNA (blunted with T4 exonuclease).
The length of the dG-tail at the 5' UTR of the human TNF cDNA is not known. The 3' UTR of the human TNF cDNA is not complete.
This plasmid contains the small t-antigen splicing signal.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727630.1.
The nucleotide sequence of the 189 bp EcoRI/SspI fragment was confirmed by sequence analysis at BCCM/GeneCorner.
Name mentioned in Müller et al. (1986) is pSVd2-hTNF (PstI)-1.
Other names of the plasmid are pSVd2-hTNF and pSV23SHuTNFwt.

Authenticity test: Restriction enzyme pattern analysed at GeneCorner: EcoRI, FspI, HindIII, NcoI and SalI. Referring to the expected fragment sizes deduced from the compiled nucleotide sequence, the presence, the length and the orientation of the hTNF insert can be confirmed, but the vector part of the plasmid seems to be larger. The plasmid seems to be about 6000 bp.
