Genecorner:

-

Cloned DNA: Human tumor necrosis factor receptor p75 cDNA (TNFR75, TNFR2, TNFRSF1B); mutant TNF-R75 M1
Promoter: Simian virus 40 early promoter (SV40 early)
Ribosome
binding site:
-
Terminator: Simian virus 40 polyadenylation signal (SV40 polyA)
Selection marker: Ampicillin (amp)
Replicon: Escherichia coli plasmid pMB1 origin
Simian virus 40 bidirectional origin (SV40)
Host range: Escherichia coli
Mammalian cells; SV40 permissive cells
Parental clone: pSV25S-HTNFR75
Further information: The plasmid was constructed as follows: the unique BstXI restriction site of pSV25S-HTNFR75 was cut, treated with T4 polymerase and a XbaI linker was inserted.
Sequencing results revealed that two linkers were inserted and that an extra C residue of the BstXI site was removed, resulting in the creation of an in frame termination codon at 5 codons downstream of the insert.
The plasmid contains the SV40 small t-antigen splicing signal.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727185.1.
The nucleotide sequence of the hTNFR75 cDNA corresponds with the EMBL Nucleotide Sequence Database (Release 37; Accession number S63368) completed with the 5' UTR, the signal sequence and the initial 15 codons of the mature hTNFR75 gene obtained from the EMBL Nucleotide Sequence Database (Release 42; Accession number M32315). However, the hTNFR75 coding sequence contains one silent mutation (W. Lesslauer: personal communication).
