Further information: | The plasmid was constructed as follows: 1) The SalI-MstII fragment of pSV51cE6Hy3 was removed; 2) After filling in the SalI and MstII sticky ends with Klenow DNA polymerase, at least 2 multi-stop-linkers 'CTTAGTTAACTAAG' were added, repairing the SalI site; the succession of the two linkers resulted in the fomation of a second HindIII site. The exact number of the multi-stop-linkers is unknown: certainly more than one, because an additional HindIII site was found. After translation, E6Hy3f2 contains the first 27 amino acids of the original CH2 domain followed by 4 amino acids derived from the cloning strategy and the multi-stop-linker. The chimeric CH1 domain contains 60 amino acids of mouse (E6H) and 37 amino acids of human (hIG3) origin. The nucleotide sequence of the pOMMB part (located between nucleotide 3394 and 3126; taken from 'GENBANK Release 55; Locus : HUMIGHAF') differs in 1 nucleotide from our own determined E6Hy3 nucleotide sequence, but there are no differences in amino acid sequence. |
