Further information: | pSV71BlahCH3 was constructed to screen for interacting CH3 domains (minibodies). The plasmid contains the mutated ampicillin resistance gene in which the termination codon has been replaced by GAG, fused in frame to the CH3 domain of the human immunoglobulin γ3 heavy chain (BlahCH3). The complete fusion gene is placed under control of the SV40 late promoter. The ampicillin resistance gene lacks its own promoter. Nevertheless, the transformants grow on ampicillin selective medium. β-lactamase is also enzymatically active after expression in COS-1 cells. pSV71BlahCH3 differs from pSV71BlahCH3.1 in the CH3 domain originating from the human immunoglobulin γ3 and γ1 respectively. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727301.1. |
