| Further information: | The plasmid was constructed as follows: 1) The rat PDI gene was isolated as an ApaI (blunted with T4 DNA polymerase) - SmaI fragment from ppdi100 and XbaI linkers ('CCTCTAGAGG') were added; 2) Subsequently, the rat PDI gene was cloned as an XbaI fragment into the XbaI opened pSV71, in the sense orientation relative to the SV40 late promoter. pSV71rPDI1 was used for the production of enzymatically active rat protein disulfide isomerase in COS-1 cells. As this plasmid contains duplicated regions of SV40 DNA, one should be aware of the possibility of deletions due to homologous recombination. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727426.1. The nucleotide sequence of the rPDI cDNA corresponds with the EMBL Nucleotide Sequence Database (Release 36; Accession number X02918). The nucleotide sequences at the 5' end and at the 3' end of the rPDI cDNA insert are not exactly known. |
