| Further information: | The plasmid was constructed as follows: the HindIII (blunted with Klenow DNA polymerase)- SstI fragment of pBLhIL6LUC (containing the luciferase gene) was inserted between the SmaI and SstI sites of pSVL. The SV40 late transcripts have multiple cap sites. The most important transcription initiation site is at nucleotide position 853 (corresponds to L325 in SV40). The plasmid gives luciferase expression after transfection into COS cells. |
