Further information: | The plasmid was constructed by cloning the synthetic HA-L40-P2A-VSV-27a-P2A-Etag cassette into the pUC19 vector. The polycistronic construct [BamHI]-HA-L40-P2A-VSV-27a-P2A-Etag-[EcoRI] (PolyCisRibo) is meant to be used for ubiquitin-replacement constructs, where mutated/tagged ubiquitins are cloned upstream of the PolyCisRibo and a selectable marker downstream of the PolyCisRibo. HA, VSV and E-tags are used to monitor processing and maturation of the construct. P2A is a self-cleaving peptide, fulfilling the same role as an IRES, but has a much shorter sequence. |
