Cloned DNA: Influenza A virus (A/Victoria/3/1975(H3N2)) haemagglutinin cDNA (HA, IVHA); mutated sequence, N-terminal anchor free
Promoter: Escherichia coli lac operon promoter
Ribosome
binding site:
-
Terminator: -
Selection marker: Ampicillin (amp)
Replicon: Escherichia coli plasmid pMB1 origin
Host range: Escherichia coli
Parental clone: pUC18; pSV23IVHAsm
Further information: The plasmid was constructed by inserting the SalI-BamHI fragment of pSV23IVHAsm, containing the A/Victoria/3/75 influenza haemagglutinin cDNA encoding a mutated, secreted protein (IVHAsm), into the SalI-BamHI opened pUC18 vector.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727288.1.
EMBL Accession number: LT727288.1, view at EMBL, GenBank, DDBJ
