Further information: | The plasmid was constructed by cloning the HA-tagged synthetic human DRD4-2 coding sequence, lacking codons 290 up to and including 295, into the pcDNA3 vector. The synthetic human DRD4-2 coding sequence was designed by Snyder & Sakmar (unpublished). It was derived from D4 receptor variant sequences presented in GenBank Accession Number L12398 and contains a direct repeat (D4-2) of a 48 basepair sequence in the putative third cytoplasmic loop. The sequence has been altered by inserting new restriction sites while maintaining the original amino acid sequence. Codons 290 up to and including 295 have been removed from the coding sequence. The nucleotide sequence of the synthetic human DRD4-2 coding sequence corresponds with the EMBL Nucleotide Sequence Database accession number AF119328.1. Other names of the plasmid are pcDNA3HAD4.2(∆290-295) and pcDNA3-HA-hDRD4-2(d290-295). |
