| Further information: | The plasmid was constructed as follows: the BamHI-BanII (blunted with T4 DNA exonuclease) fragment of pUC18gMIL6, containing the mouse IL6 promoter, was inserted between the BamHI-BglII (filled-in with Klenow DNA polymerase) sites of pBLCAT5, hereby replacing the TK promoter. pBLmIL6CAT contains the SV40 small t-antigen splicing signal. pBLmIL6CAT contains also three SV40 polyadenylation segments. Hence, the possibility might exist that the plasmid could loose information by homologous recombination. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727418.1. The nucleotide sequence of the genomic mouse interleukin 6 DNA corresponds with the Genbank accession number M20572.1. |
