Further information: This plasmid was constructed by inserting the EcoRI fragment from the 88.YH vector, containing the human IL1B prosequence cDNA, into the EcoRI opened pCAGGS vector.
pCAGGS-pIL-1B is useful for highly efficient expression of the human IL1β prosequence under the control of the AG promoter and the human CMV-IE enhancer in various mammalian cells.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727210.1.
The nucleotide sequence of the human IL1B insert corresponds with the EMBL Nucleotide Sequence Database accession number M15330.1.
Since the nucleotide sequence at the 3' end of the human IL1B insert could not be completely traced back, there is uncertainty as to the cutting frequency of the restriction enzymes, except for the sites that were analysed in the authenticity test.
Other name of the plasmid are pcaggshumanpIL1β#12a and pCAGGS-pro-IL-1beta.
Name mentioned in Kersse et al (2011) is pCAGGS-pro-IL-1B.
