Further information: | The plasmid was constructed by ligation of three fragments: 1) the XhoI-NruI fragment from pBRphoAE6L, containing on the one hand the Escherichia coli alkaline phosphatase A (phoA) signal sequence fused in phase to cDNA encoding the light chain of the mouse anti-hPLAP monoclonal antibody E6(IgG2b,κ), E6L, missing the leader signal sequence, and on the other hand the tac promoter; 2) the large NruI-MluI fragment from pDMI,1, containing the neomycin resistance gene, the p15A origin of replication, the lacI(q) repressor promoter and the 5' part of the lacI repressor gene; 3) the MluI-SalI fragment from pDMI,1, containing the remaining part of the lacI repressor gene. The plasmid is used for co-expression with plasmids encoding the heavy chain of the mouse anti-hPLAP monoclonal antibody E6(IgG2b,κ), E6H. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727563.1. Name mentioned in Flamez et al. (1995) is pDMI,1E6L. Other name of the plasmid is pDMI,1phoAE6L. |
