Further information: The plasmid was constructed by inserting a 756 bp BamHI-XbaI fragment, containing the intermediate domain (ID, encoding amino acids 304-552), obtained after N-terminal deletion of the kinase domain and C-terminal deletion of the death domain, of the mouse Ripk1 cDNA, between the BamHI and the XbaI site of pEF1-mRIP-V5/His. As a result, mouse Ripk1-ID was fused in phase to the C-terminal V5 epitope and histidine tag.
pEF1-mRIP1-ID-V5-His is designed for high-level expression of mouse Ripk1-ID under control of the human EF1α promoter in mammalian cells.
There is uncertainty about the presence of a second enhancer in the SV40 early promoter. The absence of a second enhancer in this promoter may cause bacterial leakage expression of the Tn5 neomycin resistance gene.
This would cause transformed E. coli cells to be resistant to kanamycin, although growth should be reduced compared to growth on medium containing ampicillin.
The nucleotide sequence of the mouse Ripk1 cDNA corresponds with the EMBL Nucleotide Sequence Database accession number BC058162.1.
Other names of the plasmid are pEF1-mRIP1-ID-V5/His and pEF1-mRIP-ID-V5/His.
