Further information: The plasmid was constructed by cloning the human TRAP1 cDNA as an EcoRI-BamHI fragment into the pEGFP-N1 vector. The human TRAP1 cDNA is lacking the first 58 codons, thereby eliminating its mitochondrial localisation signal. The EGFP cds is fused to the C-terminus of TRAP1.
The depositor has tested this construct in transient transfections: TRAP1 goes to the cytoplasm as judged by fluorescence microscopy and cellular fractionation.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT726828.1.
The N-terminal region of the human TRAP1 cds, downstream from the pMB1 ori, and the C-terminal region of the human TRAP1 cds, upstream of the SV40 polyA signal, were sequenced by LMBP.
The nucleotide sequence of the human TRAP1 cDNA corresponds with the EMBL Nucleotide Sequence Database accession number AF154108.1.
The nucleotide sequence of the pEGFP-N1 vector corresponds with the EMBL Nucleotide Sequence Database accession number U55762.1.
The nucleotide sequence of the EGFP DNA corresponds with the EMBL Nucleotide Sequence Database accession number U55761.1.
