Further information: | This Gateway entry vector was constructed by cloning the mouse Park2 mutated sequence N-terminally fused to the GFP coding sequence into the pENTR3C vector. The mouse Park2 coding sequence contains 4 mutations: the G nucleotide on position 96 of the wild type CDS is replaced by a T, resulting in a K32N amino acid change; there is an additional GCA codon between nucleotide positions 411 and 412, introducing an additional alanine amino acid; the C nucleotide on position 1140 of the wild type CDS is replaced by a T, but does not result in an amino acid change; the G nucleotide on position 1192 of the wild type CDS is replaced by a C, resulting in an E398Q amino acid change. The nucleotide sequence of the wild type mouse Park2 coding sequence corresponds with the Genbank accession number NM_016694.4. Other name of the plasmid is pENTR3C-GFP-mParkin+stop (with mutations). |
