Further information: The plasmid was constructed as follows: The ans1 sequence was isolated as a 3.5 kb EcoRI fragment from pDJB2 and cloned into the unique EcoRI site of pSal43 (orientation not known).
The 1.8 kb PvuII-PstI fragment of the ans1 sequence enhances the efficiency of transformation of this plasmid (4 to 10 times better as compared to pSal43).
This plasmid also contains a 2.6 kb SalI fragment of Aspergillus nidulans DNA, containing the structural gene for ornithine carbamoyl transferase (argB).
Some of the unique restriction sites mentioned are possibly not unique, because the nucleotide sequence is not completely known.
Other name of the plasmid is pFANS.
