Further information: | The plasmid was constructed by inserting the NotI-BglII fragment from pCAGGS-E-mABIN-2m2AQ, containing the mutated mouse A20-binding inhibitor of NF-κB activation 2 cDNA (mABIN-2), into the NotI-BglII opened pGAD424-mABIN-2 vector. As a result, the coding sequence of the mutated mABIN-2 cDNA is fused in phase to the Saccharomyces cerevisiae GAL4 activation domain (GALad). The fusion protein will be targeted to the nucleus by the nuclear localization signal of the SV40 large T-antigen that has been added to the GAL4 activation domain sequence. As compared to the wt mABIN-2 cDNA, the codons at position 269 (Leu) and 270 (Glu) were replaced by an Ala and a Gln codon respectively. The nucleotide sequence of the mABIN-2 cDNA corresponds with the EMBL Nucleotide Sequence Database accession number AJ304865.1. Other name of the plasmid is pGAD424-mABIN-2(LE269AQ). |
