Further information: The plasmid was constructed by inserting the CaBAX cDNA as a PstI/SmaI fragment between the PstI and StuI sites of pGAL1ACT1LUC. To facilitate the recognition of the AUG codon, a purine nucleotide (A) was introduced at position -3 (Kozak).
pGAL1P:synthCaBAX is used for expression of CaBAX after linearisation with Bpu1102I (EspI) and integration in the Candida genome.
As compared to the sequence of the pMB1 origin of pBR322, the sequence file of pGAL1P:synthCaBAX, which was obtained from the depositor, contains a ClaI site in the pMB1 region. This site has not been verified by BCCM/GeneCorner.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727209.1.
The nucleotide sequence of the Candida Wh11 terminator corresponds with the EMBL Nucleotide Sequence Database accession number S65451.1.
The nucleotide sequence of the CaURA3 DNA corresponds with the EMBL Nucleotide Sequence Database accession number AF109400.1. This was confirmed by the sequence analysis results of BCCM/GeneCorner.
